Yagami Light es un estudiante aparentemente normal, pero en su fuero interno es un brillante sociópata, sádico y megalómano que detesta el crimen y la maldad que existe en el mundo, y sueña con un entorno utópico libre de tales lacras. Su vida cambia radicalmente cuando encuentra en el suelo un misterioso cuaderno en cuya portada se lee DEATH NOTE (Cuaderno de muerte). Dentro del cuaderno hay instrucciones sobre su uso, donde destaca el hecho de poder matar a una persona (siempre y cuando se conozca el rostro de ella) de un ataque al corazón 40 segundos déspues de escribir el nombre de aquella persona en la Death Note. Light se mantiene escéptico de la autenticidad del cuaderno, pero después de probarlo en dos ocasiones, se da cuenta que su poder es real. Pasados algunos días lo visita un shinigami llamado Ryuk. Los shinigamis son seres de otra dimensión que usan los Death Note para matar personas y así prolongar su propia existencia. Ryuk le explica a Light que el Death Note que encontró le pertenece, pero cuando se extravia, automáticamente pasa a ser posesión de la persona que la encuentre, hasta que ésta última muera. A su vez Light le cuenta a Ryuk que su objetivo es matar a los criminales, para así limpiar al mundo de la maldad y convertirse en el dios de un mundo nuevo, libre de crimen.
Pronto, las numerosas muertes sin explicación atraen la atención de la policía, la cual pide ayuda a un misterioso detective mundialmente conocido como L. L se da cuenta que Kira (apodo con el cual se conoce al asesino) puede matar a las personas con sólo conocer su nombre y su rostro. Ambos empiezan una pelea de ingenio y astucia, enfrentándose a una serie de situaciones, siempre con el fin de descubrir la identidad del otro y de demostrar quién es la justicia.
Misa Amane
Es una famosa idol de Japón que siente devoción por Kira, pues fue Yagami quien terminó dándole muerte al ladrón que asesinó a sus padres. Misa posee otro "Death Note" aparte del que tiene Light. Misa intenta por todos los medios contactarse con Kira. Cuando se encuentra con Yagami, se enamora profundamente de él. Decide luego, por amor, ayudar a Light en su misión de limpiar el mundo. Misa es bastante ingenua, pero no tan estúpida como todos piensan ni tampoco sirviente de Kira, pues puede ser muy imperante cuando se trata de evitar que Light salga con otras chicas. Misa obtuvo su Death Note gracias a que Jealous (en inglés significa celoso), un shinigami, se enamoró de ella y la salvó asesinando al acosador que estaba destinado a quitarle la vida a Misa. Debido a este hecho, Jealous murió. Así que Rem, el shinigami que normalmente acompaña a Misa, decide entregarle ese Death Note a ella, explicándole que, al sacrificarse Jealous por ella, el Death Note de Jealous por regla pasa a ser ahora suyo. Físicamente es muy reconocida por su belleza, su cabello es rubio (pelirrojo en el manga original), es delgada, mide 152 cm y pesa 36 kg
